GDSLock Hotstart Taq Polymerase is a ligand-modified hot-start DNA polymerase utilizing covalent chemical blocking technology. Through reversible covalent binding of specific small-molecule ligands to the enzyme's active center, the polymerase activity is blocked by ≥99% at ambient temperature, completely eliminating unintended extension caused by non-specific primer annealing or primer-dimer formation during reaction setup and thermal ramping.
Unlike antibody-based hot-start enzymes, this product is animal-origin-free, completely avoiding the potential introduction of animal-derived protein contaminants associated with antibody-based methods. It is the ideal choice for IVD kit manufacturing and forensic DNA analysis.
Upon heating to 95°C, the ligand molecules are slowly released and dissociated within 1-5 minutes, with enzymatic activity gradually restored to the level of native Taq polymerase. This "time-released thermal activation" mechanism ensures stringent low-temperature blocking while preventing potential substrate competition caused by instantaneous release, thereby further enhancing amplification uniformity.
GDSLock Hotstart Taq Polymerase possesses 5’-3’ polymerase activity, lacks 3’-5’ exonuclease activity, and produces amplicons with 3’-dA overhangs.
